About RLY-2608 RLY-2608 is the lead program in Relay Therapeutics' efforts to discover and develop mutant selective inhibitors of PI3Kα, the most frequently mutated kinase in all cancers, with oncogenic mutations detected in about 14% of patients with solid tumors. RLY-2608 has the potential, if approved, to address more than 300,000 patients per year in the United States, one of the largest patient populations for a precision oncology medicine. Traditionally, the development of PI3Kα inhibitors has focused on the active, or orthosteric, site. The therapeutic index of orthosteric inhibitors is limited by the lack of clinically meaningful selectivity for mutant versus wild-type (WT) PI3Kα and off-isoform activity. Toxicity related to inhibition of WT PI3Kα and other PI3K isoforms results in sub-optimal inhibition of mutant PI3Kα with reductions in dose intensity and frequent discontinuation. The Dynamo® platform enabled the discovery of RLY-2608, the first known allosteric, pan-mutant, and isoform-selective PI3Kα inhibitor, designed to overcome these limitations. Relay Therapeutics solved the full-length cryo-EM structure of PI3Kα, performed computational long time-scale molecular dynamic simulations to elucidate conformational differences between WT and mutant PI3Kα, and leveraged these insights to support the design of RLY-2608. RLY-2608 is currently being evaluated in a first-in-human trial designed to treat patients with advanced solid tumors with a PIK3CA (PI3Kα) mutation. The Associated Press created this story using technology provided by Data Skrive and data from Sportradar .WASHINGTON, Dec 9 (Reuters) - U.S. government agencies will hold a classified briefing for the House of Representatives on Tuesday on China's alleged efforts known as Salt Typhoon to infiltrate American telecommunications companies and steal data about U.S. calls, officials said on Monday. The FBI, the Office of the Director of National Intelligence, Federal Communications Commission Chair Jessica Rosenworcel, the National Security Council and the Cybersecurity and Infrastructure Security Agency are set to take part in the 2:15 p.m. ET briefing for all House lawmakers after holding a similar closed-door briefing last week for senators. The White House last week said at least eight telecommunications and telecom infrastructure firms in the United States had been impacted and a large number of Americans' metadata has been stolen in the sweeping cyber espionage campaign. The White House did not immediately comment. Democratic Senator Ron Wyden told reporters after the briefing last week he was working to draft legislation on this issue, while Senator Bob Casey said he had "great concern" about the breach and added it may not be until next year before Congress can address the issue. Separately, a Senate Commerce subcommittee will hold a Wednesday hearing on Salt Typhoon and how "security threats pose risks to our communications networks, and review best practices." The hearing will include Competitive Carriers Association CEO Tim Donovan. Chinese officials have previously described the allegations as disinformation and said Beijing "firmly opposes and combats cyber attacks and cyber theft in all forms." There is growing concern about the size and scope of the reported Chinese hacking into U.S. telecommunications networks and questions about when companies and the government can assure Americans over the matter. "The extent and depth and breadth of Chinese hacking is absolutely mind-boggling - that we would permit as much as has happened in just the last year is terrifying," said Senator Richard Blumenthal. U.S. officials have previously alleged the hackers targeted Verizon (VZ.N) , opens new tab , AT&T (T.N) , opens new tab , Lumen (LUMN.N) , opens new tab and others, and stole telephone audio intercepts along with a large tranche of call record data. Tech baron Elon Musk has likened the ABC to Russian state media after the national broadcaster’s head said the likes of podcaster Joe Rogan “prey on people’s vulnerabilities” and “contribute to uncertainty in society”. Mr Musk, like Mr Rogan, has positioned himself as a free speech absolutist and mustered enormous global support for promoting unfettered debate. Taking to his social media platform X, Mr Musk reposted Mr Rogan’s reaction to ABC chair Kim Williams’ remarks overnight, writing: “From the head of Australian government-funded media, their Pravda.” Pravda is a state-owned Russian newspaper that has served as a major mouthpiece for the Kremlin for more than 100 years. Mr Rogan himself reposted a clip of Mr Williams’ comments along with a two-word reaction: “LOL WUT.” While many view both Mr Rogan’s podcast and Mr Musk’s X as allowing ideas to flow freely, they have also been accused of fuelling falsehoods and promoting harmful narratives. Mr Williams made the remarks when asked about the enormous influence of podcasters during a National Press Club appearance on Wednesday. “I think people like Mr Rogan prey on people’s vulnerabilities,” he said. “They prey on fear. They prey on anxiety. They prey on all of the elements that contribute to uncertainty in society. “And they entrepreneur fantasy outcomes and conspiracy outcomes as being a normal part of social narrative.” He said he found it “deeply repulsive” and that “to think that someone has such remarkable power in the United States is something that I look at in disbelief.” “I’m also absolutely in dismay that this can be a source of public entertainment when it’s really treating the public as plunder for purposes that are really quite malevolent,” he said. Mr Musk, the world’s richest person, has had a number of fiery encounters with the Albanese government over free speech. He will head Donald Trump’s new Department of Government Efficiency in the US president-elect’s incoming administration.By GABRIELA SÁ PESSOA and MAURICIO SAVARESE, Associated Press SAO PAULO (AP) — Brazil’s federal police said Thursday they indicted former President Jair Bolsonaro and 36 other people for allegedly attempting a coup to keep him in office after his defeat in the 2022 elections. Police said their findings were being delivered Thursday to Brazil’s Supreme Court, which must decide whether to refer them to Prosecutor-General Paulo Gonet, who will either formally charge Bolsonaro and put him on trial, or toss the investigation. The former right-wing president has denied all claims he tried to stay in office after his narrow electoral defeat in 2022 to his rival, leftist President Luiz Inácio Lula da Silva. Bolsonaro has faced a series of legal threats since then. Police said in a brief statement that the Supreme Court had agreed to reveal the names of all 37 people who were indicted “to avoid the dissemination of incorrect news.” The 700-page police document likely will take several days for the court to review, Supreme Court justice Alexandre de Moraes said. Dozens of former and current Bolsonaro aides also were indicted, including Gen. Walter Braga Netto, who was his running mate in the 2022 campaign; former Army commander Gen. Paulo Sérgio Nogueira de Oliveira; Valdemar Costa Neto, the chairman of Bolsonaro’s Liberal Party; and his veteran former adviser, Gen. Augusto Heleno. The investigation started last year. On Tuesday, four military men and one federal police agent were arrested as part of the same probe . Other investigations focus on Bolosnaro’s potential roles in smuggling diamond jewelry into Brazil without properly declaring them, and in directing a subordinate to falsify his and others’ COVID-19 vaccination statuses. Bolsonaro has denied any involvement in either. Another probe found that he had abused his authority to cast doubt on the country’s voting system, and judges barred him from running again until 2030. The far-reaching investigations have weakened Bolsonaro’s status as a leader of Brazil’s right wing, said Carlos Melo, a political science professor at Insper University in Sao Paulo. “Bolsonaro is already barred from running in the 2026 elections,” Melo told the The Associated Press. “And if he is convicted he could also be jailed by then. To avoid being behind bars, he will have to convince Supreme Court justices that he has nothing to do with a plot that involves dozens of his aids. That’s a very tall order,” Melo said. On Tuesday, the federal police arrested four military and a federal police officer accused of plotting to overthrow the government following the 2022 elections, including alleged plans to kill Lula and other top officials.None
The condemnation came as the House of Lords debated regulations paving the way for a scheme which would require animal lovers on the British mainland to have documentation in order to visit Northern Ireland. Critics view the move as further evidence of Northern Ireland still having to follow EU rules post-Brexit and being treated differently from the rest of the UK – a major source of contention to the unionist community. The paperwork, which will be free to apply for, includes a declaration that the owner will not travel onwards to Ireland or another EU country with their pet or assistance dog. Animals will have to be microchipped and have their own individual pet travel document, which will be valid for its lifetime. Northern Ireland residents returning after a stay in Great Britain with their pet or assistance dog will not need a travel document. The scheme is being introduced under the Windsor Framework, a revised deal for Northern Ireland’s post-Brexit trading arrangements aimed at tackling issues caused by the protocol. Raising her concerns in Parliament, Baroness Hoey, a Northern Irish Brexit supporter and former Labour MP, said: “These regulations are in effect about a new aspect of the Irish Sea border that has not had expression until this point because of the grace periods.” She added: “The experience of visiting Northern Ireland with your pet dog or cat, or even a ferret, will be made to feel like a visit to a foreign country. Lady Hoey went on: “This could spell the end of holiday trips for pet owners from GB to NI and then on to the Republic, when they want to explore both Northern Ireland and the Republic. “If they have a pet passport, they will have renounced their right to go to the Republic. That makes the border more of an obstruction than having border control posts on it, because at least in that eventuality, you could still cross over it.” Rejecting claims it was a result of the UK leaving the EU, she said: “The reality is that this is happening precisely because Northern Ireland has not got Brexit. “As we say repeatedly, it is still subject to EU rules and the EU could change the rules overnight.” Former DUP deputy leader Lord Dodds of Duncairn said: “Every one of the statutory instruments that come forward under the Windsor Framework must be properly debated, because these laws are being brought forward to implement what a foreign jurisdiction has decided should be the law of the United Kingdom. “In the 21st century, we should not accept colonial rule. We abolished it elsewhere. We believe it should not be tolerated for one second. People should have the democratic right to decide their laws for themselves, in their interests.” He added: “The ridiculous part about this debate is that we are having to debate European laws regulating the movement of pet animals owned by British citizens between one part of the United Kingdom and another. That is an outrage.” Lord Dodds went on: “As I said, there will be hundreds, thousands more of these regulations, in all areas, affecting the daily lives of people in Northern Ireland. They all add up to a grievous assault on Northern Ireland’s constitutional position.” But former leader of the Social Democratic and Labour Party (SDLP) Baroness Ritchie of Downpatrick said: “I support the Windsor Framework because it is a necessary legal device to deal with the complexities that were presented to us in Ireland, north and south, on the issue of Brexit. “We need a pragmatic solution rather than choosing to have political contests and duels simply for the sake of them.” Introducing the regulations, environment minister Baroness Hayman of Ulloch said: “This scheme will simplify the requirements associated with moving pet dogs, cats and ferrets from Great Britain to Northern Ireland significantly. “It replaces single-use animal health certificates with a free-of-charge lifelong travel document and removes the need for costly pet health treatments. “Pet owners who travel frequently with their pets, or those who rely on the services of an assistance dog to travel independently, will benefit substantially from this change in approach.” However, she acknowledged the concerns raised by peers and promised to continue engagement with them. Why some brewing companies are producing more hop-forward ales and light-bodied lagers
Berlin confirmed plans to reform its legal framework make it a clear criminal offence to “facilitate the smuggling of migrants to the UK” as part of the agreement, the Home Office said. The Home Office said the move would give German prosecutors more tools to tackle the supply and storage of dangerous small boats. Both countries will also commit to exchange information that may help to remove migrant-smuggling content from social media platforms and tackle end-to-end routes of criminal smuggling networks as part of the deal. It comes ahead of the UK and Germany hosting the so-called Calais Group in London, which sees ministers and police from the two countries, alongside France, Belgium and the Netherlands, gather to discuss migration in Europe. Delegates are expected to agree a detailed plan to tackle people-smuggling gangs in 2025 at the meeting on Tuesday. Home Secretary Yvette Cooper said: “For too long organised criminal gangs have been exploiting vulnerable people, undermining border security in the UK and across Europe while putting thousands of lives at risk. “We are clear that this cannot go on. “Germany is already a key partner in our efforts to crack down on migrant smuggling, but there is always more we can do together. “Our new joint action plan with deliver a strengthened partnership with Germany, boosting our respective border security as we work to fix the foundations, and ultimately saving lives.” Nancy Faeser, German federal minister of the interior said: “We are now stepping up our joint action to fight the brutal activities of international smugglers. “This is at the core of our joint action plan that we have agreed in London. “I am very grateful to my British counterpart Yvette Cooper that we were able to reach this important agreement. “It will help us end the inhumane activities of criminal migrant smuggling organisations. “By cramming people into inflatable boats under threats of violence and sending them across the Channel, these organisations put human lives at risk. “Many of these crimes are planned in Germany. “Together, we are now countering this unscrupulous business with even more resolve. “This includes maintaining a high investigative pressure, exchanging information between our security authorities as best as possible, and persistently investigating financial flows to identify the criminals operating behind the scenes.”Brookfield Asset Management Ltd. Luigi Mangione has been arrested in an investigation into UnitedHealthcare CEO Brian Thompson's killing. Mangione, an Ivy League graduate, left an online trail before his arrest at McDonald's. He founded an app, talked about Artificial Intelligence on X, and read The Unabomber Manifesto. Luigi Mangione, a 26-year-old Ivy League graduate, left a vast online trail before police in Pennsylvania picked him Monday as a "person of interest" in the Manhattan killing of UnitedHealthcare CEO Brian Thompson . He was arrested on local gun charges, but New York Police Department officials say he's expected to be extradited to New York, where he'll face additional charges. Here's what to know about Mangione: Mangione attended elite schools Mangione graduated from the University of Pennsylvania in 2020. He achieved a Bachelor of Science in Engineering with a major in computer science and a minor in mathematics. He also received a Master of Science in Engineering the same year with a major in computer and information science, a university spokesperson told Business Insider. Before that, he attended Gilman School, an elite all-boys preparatory school based in Baltimore. His yearbook entry, obtained by BI, says he was involved in robotics and Model UN. He wrote on his LinkedIn account that he was the valedictorian of his high school class. A spokeswoman for Gilman didn't immediately return a voicemail from BI. He favorably reviewed The Unabomber Manifesto On Goodreads, Mangione reviewed Ted Kaczynski's "Industrial Society and Its Future" book, also known as The Unabomber Manifesto, in early 2024. He gave it four out of five stars. "He was a violent individual — rightfully imprisoned — who maimed innocent people," Mangione wrote. "While these actions tend to be characterized as those of a crazy luddite, however, they are more accurately seen as those of an extreme political revolutionary." Mangione's review of the manifesto also quoted another online comment about the book, which appears to have originated on Reddit, praising the use of violence "when all other forms of communication fail." "'Violence never solved anything' is a statement uttered by cowards and predators,'" Mangione quoted. Online breadcrumbs suggest he dealt with back pain At the top of Mangione's profile on X — formerly Twitter — was a triptych of three images: a photo of himself, smiling, shirtless on a mountain ridge; a Pokemon; and an x-ray with four pins or screws visible in the lower back. Related stories Some of the books reviewed on Mangione's Goodreads account related to health and healing back pain, including "Back Mechanic: The Secrets to a Healthy Spine Your Doctor Isn't Telling You" and "Crooked: Outwitting the Back Pain Industry and Getting on the Road to Recovery." The Pokemon featured in his X cover image, next to an apparent spinal X-ray, is Breloom. In the Pokemon games, it has special healing abilities. He founded an app and worked in tech While still in high school, in 2015, Mangione founded a company called AppRoar Studios. AppRoar released an iPhone game called Pivot Plane that is no longer available, but a reviewer in 2015 called it "a fun little arcade game brought to you by 3 high school juniors." He lived in a coliving space in Hawaii as recently as 2023. He posed for photos indicating he participated in Greek life at the University of Pennsylvania. The fraternity chapter represented in his photos couldn't be reached for comment. He cofounded a video game design club at the University of Pennsylvania, according to a blog post on the University of Pennsylvania's website that was removed Monday. Stephen Lane, a professor of video game design at the Ivy League university, who did not advise the club, told BI that "the fact he took the initiative and started something from nothing, that means at least in the context of Penn, that's a pretty good thing." However, the shooting of Thompson, Lane added, was "obviously not a good thing." According to his LinkedIn page, Mangione worked as a data engineer at the vehicle shopping company TrueCar starting in 2020. A TrueCar spokesperson told BI that Mangione has not worked for the company since 2023. He was previously cited for trespassing Mangione had at least one encounter with the legal system before his Monday arrest. Hawaiian court records indicate that he was cited in 2023 for entering a forbidden area of a state park. Mangione appears to have paid a $100 fine to resolve the matter. Mangione was interested in AI On his X account, Mangione posted and amplified posts about technological advances like artificial intelligence. He also posted about fitness and healthy living. He frequently reposted posts by the writer Tim Urban and commentator Jonathan Haidt about the promise and perils of technology. He also appeared to be a fan of Michael Pollan, known for his writing about food and ethics, and lab-grown meat. On Goodreads, he praised Urban's book "What's Our Problem?: A Self-Help Book for Societies," describing it as "one of the most important philosophical texts of the early 21st century." Urban posted to X on Monday: "Very much not the point of the book."
Building larger schools could save taxpayers up to $1.5 billion as Victoria is projected to need almost 1000 new kindergartens and government schools in the next decade to keep up with demand. Login or signup to continue reading The state will need about 900 new kindergartens, 60 new government schools and 20 per cent more space for TAFE by 2036, according to Infrastructure Victoria's Learning for Life report. While private and not-for-profit providers can meet some of the need for kindergartens, Infrastructure Victoria said the government must step up to fill in investment gaps. The north, west and southeast growth areas of Melbourne would benefit the most from this investment. Victoria will need 138,000 more kindergarten places by 2036 and that could cost up to $17 billion if the government delivers all 900 new facilities for three- to four-year-olds. But depending on the level of private investment, this could cost between $6.1 billion to $11 billion. Most demand for government schools will be in Melbourne's fast-growing communities with an estimated 10,000 to 17,000 enrolments by 2036 in Wyndham and Melton in the west, Whittlesea in the north and Casey in the southeast. The state should also expand existing schools in Melbourne's inner and middle suburbs and begin new education deliveries by 2030, Infrastructure Victoria chief executive Jonathan Spear said. "Our modelling shows the best way to deliver the schools we will need beyond 2026 is to build larger new schools and expand existing schools where there is land to do so," he said. Building larger new schools could cost $5.9 billion, a saving of $1.5 billion, compared with the government's historical approach to delivering school infrastructure. Strong demands for TAFE will require the government to find an extra 125,000 square metres, or 20 per cent, of space by 2030 for Melbourne's west, north and southeast growth areas.
