The highlighting process now finally works in amphibians. This has been shown in a new study published in Developmental Cell by an international EDGE consortium jointly led by the Sweeney Lab at the Institute of Science and Technology Austria (ISTA) and the Tosches Lab at Columbia University. The researchers established a new technique that uses adeno-associated viruses (AAVs) to track a frog's nervous system throughout its metamorphosis—a developmental transition from the early tadpole stages to its adult form. A breakthrough that can help usher amphibian neurobiology into a new era. Swimming vs. walking David Vijatovic and Lora Sweeney enter a laboratory full of water tanks. Vijatovic taps on one of them. Inside, a small mottled greenish-brown African clawed frog (Xenopus laevis) appears. Its limbs are prominent, gracefully maneuvering and gripping its surroundings. In another tank, tadpoles are swirling around using simple swimming motions. It is remarkable to think that one transforms into the other. "Frogs undergo metamorphosis," Sweeney says, "making them a great model organism for studying the transition between two movement modes—swimming and walking." A frog's development spans over 12 to 16 weeks, giving scientists time to study each stage. During these weeks, a frog embryo develops to a young tadpole, a tadpole with two legs, and a young froglet with four legs before reaching the adult stage. "By looking at the several stages of development, we can investigate these locomotive behaviors and the underlying changes in the nervous system," Vijatovic adds. Just like an electrical circuit: How frogs are wired An organism's nervous system is referred to as the neural circuit because it resembles an electrical circuit . "Nerve cells (neurons) are connected to other neurons, transmitting electrical information. How we behave, what we sense, and how we interact with the world are the product of the way our neurons communicate with each other within these circuits," explains Sweeney. The critical piece is how the circuit is wired. We know that neurons are connected but which neuron connects to which? Which other cells does a single cell talk to, and what messages does it convey? To learn more about this wiring, researchers have been using viruses, proven to be a powerful tool. Adeno-associated viruses (AAVs) are ideal in that regard. They are non-pathogenic while being able to infect a wide range of cell types, including neurons. AAVs can be modified to glow in bright green fluorescent colors under the microscope as they travel along neurons, whether in retrograde (backward, from the synapse toward the cell body) or anterograde (forward, from the cell body toward the synapse). In other words, AAVs can be used to illuminate the neural circuit from the broadcasting end to the receiving end or vice-versa. "This is a common technique used in neuroscience, especially in well-studied organisms like mice. For amphibians, it was thought that it could not be done," says Vijatovic. The power of scientific collaboration To make AAV labeling work in amphibians, Sweeney and Vijatovic joined forces with an international team of scientists from Maria Tosches' group at Columbia University, where the study's other two co-first authors Eliza Jaeger and Astrid Deryckere are based. The consortium also included researchers from Tel Aviv University, the University of Utah, the Scripps Research Institute, and the California Institute of Technology. The researchers put their heads together, drew expertise from each other, visited conferences, had countless Zoom calls, and came up with different perspectives and ideas. "When you start researching an organism that is not yet well understood, it is great to have a community where you can share information," says Sweeney. They screened existing AAVs to find what was suitable for amphibians and optimized the infecting strategy eventually developing a "how-to guide" for frogs and newts. Vijatovic summarizes his Ph.D. journey, "We started with young tadpoles, made our way to older tadpoles, and finally moved to juvenile and then adult frogs as well as adult newts. We tailored the tool to each life stage." Comparing frogs to humans: What this research says about us With this new technique, the scientists managed to apply AAVs for tracing neuron connections in amphibians. This will help them find out more about how the amphibian brain compares to that of mammals. Besides that, the new approach also opens doors to further analyzing neuronal development. With some of the screened AAV variants, the researchers can label progenitor cells at a specific point in time during the circuit's development and follow them to see what neurons they become. "This way, we can resolve the whole circuit by its development, see how it changes over time, and how the whole nervous system is built," Sweeney says. Although amphibians and mammals last shared a common ancestor about 360 million years ago, they share common traits. "By comparing the details of a frog's nervous system to a human's, we can see what we don't have and what we have," Sweeney continues. This knowledge can help us understand how the human nervous system became specialized over time. Electric vehicles hold the promise to further cut tailpipe emissions linked to thousands of premature deaths in the U.S. But EV sales in the U.S. face a U-turn under the new Trump administration, threatening to reverse progress on the climate and public health impacts of transportation pollution. New data from the U.S. Environmental Protection Agency released Monday shows that the country's vehicle fleet has reached its highest fuel efficiency and the lowest greenhouse gas emissions on record following a suite of federal policies supporting EV sales and placing stricter controls on auto emissions. "Manufacturers continue to innovate and are bringing technologies to market which will directly improve air quality, better protecting people's health and saving lives," EPA Administrator Michael Regan said in a statement. But President Donald Trump , who will return to office in January, has pledged to roll back emissions standards set by President Joe Biden and end the tax credits for EV purchases under the Inflation Reduction Act , or IRA. A new analysis finds that a repeal of the EV tax credit could sharply reduce EV sales. "Our calculation is that they would decrease by 27 percent," University of California , Berkeley, associate professor Joseph Shapiro told Newsweek . In a working paper published by UC Berkeley's Energy Institute at the Haas School of Business, Shapiro and colleagues explored what would have happened to EVs last year without the $7,500 credit for EVs in the IRA. They found that EV registrations—the combined sales and leases—would drop each year by more than 300,000. "It's the U.S. manufacturers who are disproportionately impacted," Shapiro added. The analysis found that because the IRA tax credits were crafted to support U.S. automakers, the elimination of the credits would only cause foreign-made EV sales to drop by about 2 percent. The potential shift in EV policy carries major implications for U.S. climate policy and for the U.S. auto industry, which has invested heavily in EV and battery manufacturing. There are also enormous—and often overlooked—consequences for public health. Continued Pollution Despite Progress This year is the 50th anniversary of the EPA's Automotive Trends Report. In that time, the agency said, American vehicles have become 99 percent cleaner, cutting pollutants like particulate matter, soot and carbon monoxide that contribute to asthma, heart problems and cancer. But despite that progress, the EPA said, pollution from cars and trucks still harms public health and accounts for nearly 17 percent of all U.S. greenhouse gas emissions. Ray Minjares is program director for heavy-duty vehicles at the nonprofit International Council on Clean Transportation, and he has 20 years of experience in environment, energy and transportation policy. Minjares said a 2019 ICCT study found that even with the long-term improvements to clean up cars and trucks, their exhaust still made up nearly 20 percent of all outdoor air pollution Americans encounter. Particulate matter and ozone from autos contributed to about 22,000 annual premature deaths in the U.S., the ICCT study found, and the social cost of that health burden was about $210 billion. Minjares said that even with increased fuel efficiency and better emissions controls, health impacts from pollution will persist due to "the inherent fallibility" of the internal combustion engine. "The way we get past that is we advance to zero-emission engines," Minjares said. "That's the way we deal with these problems and put them to bed permanently." Internationally, the toll from vehicle pollution was much higher, the study found, with 385,000 deaths attributable to transportation emission sources and approximately $1 trillion in health damages globally. Diesel engines are responsible for a disproportionate amount of the pollution, Minjares said. In the U.S., roughly three-quarters of the most damaging tailpipe emissions came from diesel vehicles, especially heavy-duty trucks and buses. "It would make sense from a public health perspective to really double down on controlling these heavy-duty diesel vehicles and, even better, electrifying them," he said. In addition to the tax credits for EVs, the IRA and the Bipartisan Infrastructure Act included generous support for electrifying trucks and school buses and developing the charging infrastructure for them. Minjares said the EPA's new emissions standards on heavy trucks will likely require more truck makers to go electric. Many manufacturers and truck fleet operators are finding that the switch to electric yields long-term cost savings in a competitive global market, he said. "If President Trump were to roll back federal rules on heavy-duty greenhouse gas standards, it would undermine their ability to compete with China and with Europe," Minjares said. A Global EV Transition Minjares said that in addition to federal incentives for EVs, state policies are also important, especially California's. California is not only one of the largest markets for auto sales, but it has also traditionally had a leadership role in regulating auto emissions. California's Democratic Governor Gavin Newsom said in a statement Monday that if the Trump administration removes the EV tax credit, his state will be "doubling down on our commitment to clean air" by providing additional support. "We're not turning back on a clean transportation future—we're going to make it more affordable for people to drive vehicles that don't pollute," Newsom said. Minjares said that type of state support, the auto industry's heavy investment in EVs and global market trends all point to a continuing transition to electric drive regardless of whether the U.S. federal support for EVs is repealed. "I don't think there's anything that President Trump can do to change the EV direction in this country," Minjares said. Shapiro agreed that EVs now have a momentum that will be hard to stop. Incoming U.S. president Donald Trump promised Monday to impose the tariffs on his first day in office in January. He said he would keep the tariffs in place until Canada and Mexico stop illegal border crossings and prevent drugs such as fentanyl from entering the U.S. Kinew said 25 per cent tariffs would mean a recession for his province and that Canada needs to show the new U.S. administration it’s serious about security and tackling the drug crisis. He said it begins with Canada’s pledge to NATO allies to spend two per cent of its gross domestic product on defence by 2032. “First and foremost, (it’s about) hitting that target of two per cent spending on defence,” said Kinew. “That gets us in the game just to be taken seriously as a security partner with the U.S. If we don’t do it, it’s going to become a trade problem.” In a video posted to social media, Alberta’s premier said Canada needs to reach its pledged commitment on defence. “If their trade partners are looking to be free riders on American security interests, that’s also going to harm the relationship,” Smith said, adding, “You also have to take seriously the asylum seekers.” Canada and the U.S. share a $188 billion trade partnership that supports countless jobs and communities across our country. Stocks drifted higher on Wall Street in midday trading Thursday, as gains in tech companies and retailers helped boost the market. The S&P 500 rose less than 0.1%. The benchmark index is coming off a three-day winning streak. The Dow Jones Industrial Average was up 19 points, or 0.1%, as of 12:32 p.m. Eastern time. The Nasdaq composite was up less than 0.1%. Trading volume was lighter than usual as U.S. markets reopened after the Christmas holiday. Chip company Broadcom rose 2.9%, Intel was up 0.7% and Apple gained 0.4%. While tech stocks overall were in the green, some heavyweights were a drag on the market. Semiconductor giant Nvidia, whose enormous valuation gives it an outsize influence on indexes, slipped 0.2%. Meta Platforms fell 0.9%, Amazon was down 0.5%, and Netflix gave up 1.4%. Health care stocks also helped lift the market. CVS Health rose 1.9% and Walgreens Boots Alliance rose 3.3% for the biggest gain among S&P 500 stocks. Several retailers also gained ground. While the jury is still out on the factors leading to the landslide victory for the MahaYuti coalition in the Maharashtra assembly elections, anger among Muslims is being directed against religious leaders, especially Maulana Sajjad Naumani, a Deobandi cleric whose appeal to Muslims to vote for Congress-led Maha Vikas Aghadi (MVA) is being blamed for the counter-mobilisation of Hindu votes, precipitating BJP-led MahaYuti's record-breaking victory. At a press conference in Mumbai on November 13, Naumani had released a list of candidates, mostly from MVA, asking Muslims to support them. He said that Muslims should vote for the candidates 'to defeat forces challenging the Constitution'. Naumani had, however, declined to answer questions from the media on whether he represented all Muslims, explaining that he had come up with the list of candidates after consulting scholars, social activists, and opinion makers. Naumani has since apologised for a statement made during the run-up to the elections where he said that Muslims who vote for MahaYuti should be socially boycotted. After MVA's drubbing in last week's elections, Muslims are asking the question raised at the press meet. Juned Shaikh, who runs a YouTube channel called F3 News with a wide viewership, represented the view of many Muslims when he asked, "Who did maulana consult before declaring support for the candidates? There are millions of Muslims in the state; did you have a meeting with them?" Shuiab Khateeb, trustee of Mumbai's Jama Masjid, who contested as a candidate in Mumbadevi for the Aazad Samaj Party (Kanshi Ram), said it was wrong for Naumani to endorse the entire list of MVA candidates. "On what ground did Naumani base his list of candidates? Did he consult the public?" asked Khateeb. "His statement created vote banks of Hindus and Muslims. Candidates should be endorsed on the basis of their work and potential, not because they belong to a particular political group. Naumani's statements created havoc in the minds of the voters," said Khateeb. Naumani, who was educated at a university in Madina, is based out of Uttar Pradesh though he has an establishment in Mumbai. Naumani probably did not know the ground realities in Maharashtra and was misled by other people, said senior Urdu journalist Saeed Hameed. "We talk about transparency by politicians and the government. However, the maulana did not have transparency about the machinery that he used to come up with the list of candidates Muslims should vote for in 288 seats," said Saeed. "Surveys that cover a big state require a lot of money. I am apprehensive of his research and his parameters." Saeed added that Naumani's endorsement of MVA candidates lent credence to accusations of 'vote jihad' hurled by BJP leaders like Kirit Somaiya. "It was a critical election; he should have been careful in choosing his words. He exceeded his limits as a maulana. He has to ultimately apologise for some of his statements. Also, a statement by a Tablighi maulana will not be accepted by Barelvis, Shias, and other sects." A statement similar to Naumani's was made by AIMIM leader Akbaruddin Owaisi in 2014 during his first political rally in Mumbai, said Shaikh. "He said something about Muslims uniting for 15 minutes to finish off their opponents. While the statement did not unite Muslims, it did unite the majority. That was a turning point," said Shaikh. Dr Azimuddin, president of Movement for Human Welfare, who had accompanied Naumani at the press meet, said that the cleric's remarks could have had only a marginal effect on the election results. "There were other factors that led to the huge win for MahaYuti. Questions about manipulation of voting machines remain unanswered. There were reports that money meant for distribution among voters were seized. The Sangh Parivar used Naumani's statements to create an elaborate door-to-door campaign. They worked hard; the MVA was confident of winning and did not match the campaigning. The maulana is not such a powerful person to influence voters. He did not make any polarising statement," said Azimuddin Some Muslims felt that Naumani did not deserve the harsh criticism. Abdul Razak Maniar, a social worker, said, "During every election, local community leaders suggest names of candidates who they think people should vote for. Naumani made up his list after doing local surveys. However, endorsing an entire list of candidates was not a sensible thing. It could have caused counter-mobilisation of Hindu votes."EAGAN, Minn.
