We had data showing that some azolato-bridged complexes inhibit AR signaling, which is extremely important for prostate cancer proliferation, in addition to the anticancer effect initiated by the DNA-binding. Therefore, this study was conducted to clarify the mechanism of AR signaling inhibition by the azolato-bridged complex, 5-H-Y," explains Dr. Hirota. The team used a variety of methods to evaluate AR dynamics and therapeutic effects in LNCaP prostate cancer cells. They utilized azolato-bridged complexes, cisplatin, and the AR antagonist KW-365 to explore their efficacy and performed cell viability, gene expression , and protein analyses. Additionally, the team employed immunofluorescence staining to visualize AR expression and evaluated apoptosis (programmed cell death), cell cycle distribution, and nuclear platinum accumulation. The results showed that 5-H-Y exhibited significantly stronger cytotoxic effects than cisplatin, with a low half-maximal inhibitory concentration for dihydrotestosterone (DHT)-induced cell proliferation. Moreover, 5-H-Y effectively suppressed the expression of AR-responsive genes, such as PSA and TMPRSS2, and induced apoptosis in AR-overexpressing cells. Immunofluorescence analysis confirmed that 5-H-Y promoted chromatin fragmentation, a hallmark of apoptosis, with greater efficacy observed at higher concentrations. Mechanistically, 5-H-Y was found to bind directly to AR and DNA through both noncovalent and covalent interactions. This binding induced conformational changes in AR, potentially disrupting its function. Additionally, 5-H-Y arrested cell cycle in the G2/M and sub-G1 phases, leading to apoptosis, particularly in AR-overexpressing cells. This multimodal mechanism of action distinguished 5-H-Y from cisplatin, which primarily targets DNA. The Bank of Canada has cut its key policy interest rate five times this year including last week when it reduced the benchmark by a half a percentage point to 3.25 per cent. MUMBAI: A 48-year-old actor is booked for allegedly molesting a 32-year-old woman after inviting her to his house under the pretext of collaborating on Tuesday. According to the police, the victim runs a small production house in her house in Goregaon, from where she posts reels and short videos on social media. She received messages on Facebook from an account with the actor’s name and photo used in its profile. “Seeing no reason as to why the actor would want to get in touch with her, she initially assumed it was a fake profile. However, she later asked the person chatting with her to get on a video call with her. To her surprise, it was the actor himself,” said an officer from Khar police station. The actor allegedly told her to meet him at his office in Khar to discuss about shooting. The woman went to the address given by him at around 5.30pm. He instructed her to come to the third floor, added the police. It was not an office but his residence. After she reached, a senior citizen opened the door. When she inquired about the actor, he called her directly to his bedroom. Upon opening the bedroom door, she found him naked. He insisted on her getting inside the bedroom. She told him she was uncomfortable, but he forcefully pulled her inside and touched her inappropriately. The woman managed to flee out of his house. “When she reached home, she received abusive messages from the actor. The next day, she approached the police to complain and an FIR was registered against the actor,” said the officer.
