At baseline, patients in the vorasidenib arm had a median age of 49 years (range, 31-75), and most were male (66.7%). Karnofsky performance status (KPS) scores included 100% (33.3%), 90% (54.2%), and 80% (12.5%). Most patients had WHO grade 2 tumors (91.7%). Histological subtypes in this arm included oligodendroglioma (54.2%) and astrocytoma (45.8%). Additionally, 1p19q status was co-deleted, intact, or not determined in 50.0%, 41.7%, and 8.3% of patients, respectively. All patients had received prior surgery. In total, 29.2% and 41.7% of patients had received prior radiation therapy and prior systemic therapy, respectively. At baseline among patients in the ivosidenib arm, the median age was 37 years (range, 19-66), and most were male (68.0%). KPS scores included 100% (44.0%), 90% (48.0%), and 80% (4.0%); One patient (4.0%) had a missing KPS score. Most patients had WHO grade 2 tumors (84.0%). Histological subtypes in this arm included oligodendroglioma (48.0%), astrocytoma (44.0%), anaplastic oligodendroglioma (4.0%), and anaplastic oligoastrocytoma (4.0%). Furthermore, 1p19q status was co-deleted, intact, or not determined in 52.0%, 36.0%, and 12.0% of patients, respectively. All patients had received prior surgery. In total, 28.0% and 56.0% of patients had received prior radiation therapy and prior systemic therapy, respectively. Previously reported findings from this phase 1 trial showed that 2-HG concentrations were reduced by 92.6% (95% credible interval [CrI], 76.1%-97.6%) and 91.1% (95% CrI, 72.0%-97.0%) in patients who received vorasidenib at 50 mg once daily or ivosidenib at 500 mg once daily, respectively. 4 However, the ratios of tumor to plasma were higher in patients who received vorasidenib vs those who received ivosidenib. Reductions in 2-HG were associated with lower tumor cell proliferation, reversal of IDH1/2 mutation–associated gene expression programs, increased DNA 5-hydroxy-methylcytosine, and increased tumor-infiltrating lymphocyte levels. The dose of vorasidenib at 50 mg once daily was associated with the most consistent mutant enzyme inhibition and the most preliminary antitumor activity. The median progression-free survival (PFS) was 41.4 months (95% CI, 9.3-NE) in the vorasidenib arm and 38.6 months (95% CI, 18.3-NE) in the ivosidenib arm. Among patients who had only undergone prior surgery, the median PFS was 55.2 months (95% CI, 2.4-NE) in the vorasidenib arm (n = 9) and 38.6 months (95% CI, 24.0-NE) in the ivosidenib arm (n = 9). Investigators reported no new safety signals and noted that the safety profiles of the agents in this analysis were consistent with those observed in previously published studies. Serious TEAEs and treatment-related serious TEAEs were seen in 41.7% and 4.2% of patients, respectively. Grade 3 or higher TEAEs, treatment-related grade 3 or higher TEAEs, and AEs of special interest were observed in 62.5%, 12.5%, and 12.5% of patients, respectively. TEAEs led to treatment discontinuation, dose reduction, and dose interruption in 4.2%, 4.2%, and 37.5% of patients, respectively. In the ivosidenib arm, any-grade TEAEs occurred in 100.0% of patients, and treatment-related TEAEs were seen in 72.0% of patients. Serious TEAEs were seen in 32.0% of patients, and no patients had treatment-related serious TEAEs. Grade 3 or higher TEAEs and treatment-related grade 3 or higher TEAEs were observed in 32.0% and 4.0% of patients, respectively. No AEs of special interest were reported. AMHERST — Perhaps the passionate followers of the UMass football team weren’t necessarily surprised when news hit last week that Director of Athletics Ryan Bamford had fired former Minutemen head coach Don Brown after a 6-28 record with the team; it was more so the timing of the move that rubbed people the wrong way. UMass had just come off its best four-quarter performance of the season – a 35-34 overtime loss to 8-2 Liberty – when Bamford gave Brown the boot, with only two games remaining on the Minutemen’s regular season schedule. Why didn’t Bamford let Brown, who emptied the tank during his third stint in Amherst (one that lasted nearly three seasons), finish out the season before sending him on his way? The timing of the firing may not have made much sense from a fan or player perspective. But the school’s athletic director determined it had to be done right then and there. When Brown was relieved of his duties, UMass became the 11th Group of 5 school to have a vacancy at the head coaching position. And as of Monday, there are now 13 openings across G5 schools – the Minutemen’s direct competitors when it comes to staffing hires and recruiting. Bamford said sticking out the rest of the 2024 campaign with Brown and then firing him would have prevented him from talking to potential coaching candidates until Dec. 1 (UMass’ last game is Nov. 30). In order for UMass to have the best chance at getting its guy, the AD said moving quickly was an unfortunate necessity. Even further, the transfer portal opens up on Dec. 9, and early signing day for high school recruits is the week prior to that. Entering the second week of December without a head coach in place would’ve put the Minutemen at a significant disadvantage in recruiting, according to Bamford. “In order to be in a coaching candidate pool and make sure that we have the appropriate conversations and be seen as in the market place for coaches and for agents, we really needed to open the job when we did,” Bamford said on Monday, a week removed from Brown’s dismissal. “We didn’t wanna miss the opportunity to position our job against other jobs in the market. That’s how college football has changed in the last five years, and it’s predominantly because the transfer portal opens up in early December. You wanna have a head coach in place to have their staff at least somewhat organized when the portal opens.” Bamford, who has been the UMass AD since 2015, said firing Brown was not an easy decision. The two have a strong relationship, but accumulating only two wins against FBS opponents in three years – none of which came against the MAC (where UMass is headed in 2025) – was a disappointment in Bamford’s eyes. Bamford and Brown met on a weekly basis throughout the season, and the AD said he clearly stated from Week 1 that there needed to be real progress shown on the field this fall or a change may be made. After winning one game in Brown’s first year and three in his second, four or five wins were expected to continue to show improvement. But instead, the Minutemen’s only two wins this year have come against FCS opponents, and they lost every single game against MAC programs (0-5). “I felt like after three years, we had enough in the body of work for Don to understand where we were going, and we needed some momentum going into next year as we go into the MAC,” Bamford said. “We just didn’t feel like we were able to turn the corner competitively, and there are a host of reasons for that. Without question, Don and his staff have worked tremendously hard to try to get this program turned around, and the program is in a better spot now than when Don and his group got here. There’s no doubt.” Because Bamford fired Brown when he did, UMass now owes him up to $1.4 million over the next two years. If Brown takes another coaching job over the next two years, UMass would pay the difference between that number and his new salary. Had Bamford waited until the end of the season to fire him, UMass would have owed him $800,000 over the next two years. “To be fair to Don, I wanted to make sure that we honored an important piece of the termination language in the contract,” Bamford said. “We recognize him coming back to UMass and trying to build a program that he really helped put on the map as an FCS program. I thanked him for that, and I felt like it was appropriate to make sure that we did it when we did to recognize his contributions to this university.” Bamford neglected to share any names or front-runners in the coaching search, but emphasized the importance of hiring someone by the first week of December. When a hire is ultimately made, it’ll be Bamford’s third head football coach hiring of his 10-year tenure at UMass. Over the course of the past decade, the Minutemen have a 21-89 football record, including an 8-54 mark of coaches he’s hired. The Don Brown hiring back in 2021 was seen as a coup at the time, although it didn’t work out. The AD acknowledged the pressure behind this hire, but support from the administration and the help of Chad Chatlos of TurnkeyZRG Executive Search leaves Bamford confident in making a splash. Chatlos and TurnkeyZRG are highly regarded in terms of college football hiring firms, and even helped conduct the NCAA in its search for a new president back in 2022 (ultimately former Massachusetts Governor Charlie Baker). It’s the first time Bamford has hired a search firm to assist him with the hiring process. When Walt Bell was hired in 2019, Bamford was assisted by a few advisors, but not a formal search firm. At the end of the day, who patrols the sidelines next year still falls on Bamford’s shoulders. “I have great confidence we’re going to find the right person for the job,” Bamford said. “We have a lot to sell. The job is a different job than it has been in the last two hiring cycles because we’re not [a football] independent anymore. That’s been an important ingredient in this coaching search, is the ability to go into the MAC to be resourced really well against MAC-competitive peers. I think agents and coaches, those in the business see this as a different job now. It was hard to overcome the nature of being an independent in the last two searches.” UMass is positioned to be one of the highest-resourced programs in the MAC next season, including staff compensation and NIL money. This release was published on openPR.1 2 Hyderabad: The enforcement officials of prohibition and excise department said there has been a 29 per cent increase in cases related to the violation of the Narcotic Drugs and Psychotropic Substances Act (NDPS Act). Prohibition & excise director VB Kamalasan Reddy, during the annual press meet, said the number of accused arrested in NDPS Act cases went up from 1,134 in 2023 to 1,840 in 2024. The total cases registered by the excise department for different illicit distillation offences saw a two percent increase. "In 2023, we registered 20,803 cases, whereas this year, excise officials registered 21,234 cases. Similarly, 12,727 persons were arrested in 2024, which is an 8 per cent increase," Kamalasan Reddy said. The officials identified 25 hot spots across Telangana and seized stock 27 crore worth ID liquor. Operation Dhoolpet , which was started by the excise department to eradicate the supply of ganja from Dhoolpet, led to the arrest of 317 persons and the seizure of 561 kg of ganja. Another area of focus for excise officials was the seizure of Non-Duty Paid Liquor (NDPL), where 53,226 litres of liquor were seized and 2,076 persons were arrested. Across Telangana, the number of under-investigation cases decreased from 84,115 cases in Jan to 46,187 cases. Keeping in view the upcoming New Year event, the excise director said that within GHMC limits, 40 teams were formed, and they will be on the lookout for violations. This is even as the state governor, Dr Peter Mbah, has been scheduled to present the much anticipated 2025 budget at the floor of the State House of Assembly on Tuesday. The developments were disclosed to the Government House correspondents at the weekend by the Commissioner for Youth and Sports, Barr Lloyd Ekweremadu, and the Commissioner for Budget and Planning, Hon Chris Ozongwu, shortly after the State Executive Council meeting which was presided over by the governor. This is the first time any state in the South East Nigeria will be hosting the prestigious festival since inception in 1973, making Enugu the premier state to set the stage open. With Enugu winning the hosting rights, the commissioner reiterated the government’s commitment to deliver a world-class event that will enhance the festival’s legacy of excellence, inclusivity, international outlook, and discovery of potential sports stars. Barr Ekweremadu attributed the emergence of Enugu state as the hosting state to Governor Mbah’s leadership strides and huge investment in the sports’ industry, adding that plans were on ground to upgrade and develop sports infrastructure in the state. “The development strides that Enugu State is recording under the leadership of Governor Peter Mbah are a determining factor in winning the hosting rights. Council noted that in order to deliver a world-class sports festival, government must invest in upgrading and developing our sports infrastructure, particularly the Nnamdi Azikiwe Stadium, and the Awgu Games Village,” he further stressed. The sports festival is expected to attract over 15000 athletes that will be participating in the events to Enugu where talents will be showcased and new ones discovered. Meanwhile, Hon Ozongwu said the draft of the 2025 Appropriation Bill was presented to the Executive Council where it was approved for presentation after rigorous evaluation and interrogation. “Both the medium term expenditure framework (MTEF) and the budget proposal were unanimously approved by the council after much deliberation, evaluation and interrogation. “This, therefore, sets the stage for His Excellency to submit the Appropriation Bill to the House of Assembly early next week. Last month, Cannon enjoyed a successful campaign at the 2024 Pan Pacific Masters Games. He was part of the Myths and Legends team, which won a gold medal in the 80+ years and a silver in the 75+ age group in basketball. Cannon played 10 games during the Pan Pacific Games from November 3 to 10. "It was a tough schedule, but I loved every minute," he said. "I'm at my happiest when I'm playing basketball." Despite usually being one of the oldest players on the team, Cannon is one of the fittest and is often seen after a game doing a few line sprints to keep his fitness up, putting his much younger teammates to shame. Cannon, who was born in West Sydney but has lived in the Illawarra for the past twenty-plus years, started his basketball Masters' campaign in Bankstown in 1994. Over the years, he has had some speed humps, most notably a knee replacement and, more recently, a shoulder replacement. Cannon is now fighting fit. His current weekly routine involves veterans basketball on Sunday and Wednesday, Walking basketball on Wednesday and social men's basketball on Thursday. The latter has no age restriction, meaning Cannon can and often does play against players as young as 18. Last year, Cannon and his Illawarra Walking Basketball team won the state title in Hills District. The achievement came 10 years after Cannon first played in an Australian Masters' basketball tournament in 2013. That same year, he coached Australian women for 60+ years to a gold medal at the World Masters in Italy. Cannon has also played in several World Masters Games, including those held in the Philippines, Guam, New Zealand and the USA. He also represented Australia at the 2017 FIMBA Basketball World Championships in Italy, finishing sixth out of 18 teams participating. Cannon also had the honour of being the Australian flag bearer. He was also the Australian flag bearer in 2019 in Finland and was part of the first Australian side to win a medal after the team finished fourth out of the 10 competing teams. Cannon represented Australia again in 2023 in Argentina. He is preparing to play for his country again when Australia heads to Switzerland in 2025. Other team members hail from Perth, Sydney, Brisbane and Adelaide. One of the players, Frank Tomasi, currently living in Perth, originally lived in Wollongong and recalls helping his father install the original backboard supports at the Snakepit basketball courts (now called courts 3 and 4) around 1965. Australian Basketball Hall of Fame member Adrian Hurley OAM is the coach of Australia's 80+ team heading to Switzerland. Hurley has previously been the head coach of the Australian Boomers, in charge of the Illawarra Hawks NBL team in 1980, and a Perth Wildcats championship-winning coach. Bob advised the 80+ years team will get together for a training camp in Wollongong with coach Adrian Hurley around mid-May next year in preparation for the 2025 Switzerland FIMBA Basketball World Championships.
